Uses of Truqap (Capivasertib)

Truqap (Capivasertib) is a targeted cancer therapy medication developed by AstraZeneca. It’s primarily used in the treatment of certain types of advanced breast cancer.

Truqap (Capivasertib) is a protein kinase B (AKT) inhibitor, used to treat:

• HR-positive, HER2-negative advanced or metastatic breast cancer, especially in patients with tumors carrying specific PIK3CA, AKT1, or PTEN mutations.
• It’s typically used in combination with fulvestrant in adults who have previously received endocrine-based therapy.

It works by blocking the AKT signaling pathway, which plays a key role in the survival and growth of cancer cells.

Precautions and Warnings

Before using Truqap, consider the following:

Genetic Testing Required: The presence of PIK3CA, AKT1, or PTEN mutations must be confirmed before starting therapy.

Blood Sugar Monitoring: Capivasertib can cause hyperglycemia; patients with diabetes or pre-diabetes should be monitored closely.

Heart Monitoring: May prolong the QT interval, increasing the risk of heart rhythm problems. ECG monitoring may be needed.

Liver Function: Monitor liver enzymes periodically.

Fertility and Pregnancy:May harm an unborn baby. Avoid use during pregnancy. Effective contraception is required during treatment and for at least 1 week after the last dose.

Breastfeeding: Not recommended during treatment and for 1 week after the last dose.

Drug Interactions: Avoid strong CYP3A4 inhibitors/inducers (e.g., ketoconazole, rifampin) as they may alter capivasertib levels.

Dosage

• Standard Dose: 400 mg orally, twice daily for 4 days on and 3 days off in a 28-day cycle.
• Taken in combination with fulvestrant.

• Diarrhea
• Nausea and vomiting
• Fatigue
• Rash
• Decreased appetite
• High blood sugar (hyperglycemia)
• Mouth sores
• Elevated liver enzymes

Serious Side Effects:

• Severe rash or allergic reactions
• QT prolongation (irregular heartbeat)
• Severe hyperglycemia or diabetic ketoacidosis
• Visual disturbances (rare)
• Infections due to immune suppression
