Truqap (Capivasertib) is a targeted cancer therapy medication developed by AstraZeneca. It’s primarily used in the treatment of certain types of advanced breast cancer.
Truqap (Capivasertib) is a protein kinase B (AKT) inhibitor, used to treat:
- HR-positive, HER2-negative advanced or metastatic breast cancer, especially in patients with tumors carrying specific PIK3CA, AKT1, or PTEN mutations.
- It’s typically used in combination with fulvestrant in adults who have previously received endocrine-based therapy.
It works by blocking the AKT signaling pathway, which plays a key role in the survival and growth of cancer cells.
Precautions and Warnings
Before using Truqap, consider the following:
Genetic Testing Required: The presence of PIK3CA, AKT1, or PTEN mutations must be confirmed before starting therapy.
Blood Sugar Monitoring: Capivasertib can cause hyperglycemia; patients with diabetes or pre-diabetes should be monitored closely.
Heart Monitoring: May prolong the QT interval, increasing the risk of heart rhythm problems. ECG monitoring may be needed.
Liver Function: Monitor liver enzymes periodically.
Fertility and Pregnancy:May harm an unborn baby. Avoid use during pregnancy. Effective contraception is required during treatment and for at least 1 week after the last dose.
Breastfeeding: Not recommended during treatment and for 1 week after the last dose.
Drug Interactions: Avoid strong CYP3A4 inhibitors/inducers (e.g., ketoconazole, rifampin) as they may alter capivasertib levels.
Dosage
- Standard Dose: 400 mg orally, twice daily for 4 days on and 3 days off in a 28-day cycle.
- Taken in combination with fulvestrant.
