Foundayo is contraindicated in people with a personal or family history of medullary thyroid carcinoma (MTC) or Multiple Endocrine Neoplasia syndrome type 2 (MEN 2), and in anyone with a known serious hypersensitivity to orforglipron or its ingredients. Tell your clinician about any history of pancreatitis, gallbladder disease, kidney problems, pregnancy plans, or planned surgery requiring anesthesia. Breastfeeding is not recommended while taking Foundayo.

Foundayo slows stomach emptying, which can change how other oral medicines are absorbed. Oral hormonal contraceptive users may need a non-oral method or a backup barrier method for 30 days after starting Foundayo and after each dose increase. Certain CYP3A4-related medicines can change Foundayo exposure, and the maximum dose is lower with strong CYP3A4 inhibitors. Do not combine Foundayo with another GLP-1 receptor agonist. People taking insulin or a sulfonylurea may need closer blood-sugar monitoring. Tell your healthcare team about every prescription medicine, over-the-counter product, and supplement you take.
